Origin and Development

One of a family of synthetic growth hormone-releasing peptides developed from work on enkephalin derivatives found to stimulate growth hormone release. Developed under the name pralmorelin and approved in Japan for diagnostic use.

Mechanism as Understood in the Literature

Agonist at the ghrelin receptor (GHS-R1a), prompting pituitary release of growth hormone. Less selective than ipamorelin: it is also associated with increases in cortisol and prolactin, and with appetite stimulation through ghrelin receptor activity.

Research Status

Human data exists, primarily from its development as a diagnostic agent and from studies of growth hormone axis function. Its use as a diagnostic tool in Japan means its acute pharmacology in humans is better characterized than for most compounds in this category.

Commonly Confused With

Frequently grouped with GHRP-6 and hexarelin, which act on the same receptor but differ in potency and in their effects on cortisol, prolactin, and appetite. Also commonly confused with GHRH analogues such as CJC-1295 and sermorelin, which act on a different receptor entirely.
